Biuret test

    When the Biuret reagent is added to a solution containing proteins or peptides, the copper ions in the reagent form coordination complexes with the nitrogen atoms of adjacent peptide bonds. This results in a violet to purple color change.

  • Negative Result (No Peptide Bonds): If there are no peptide bonds in the solution, the Biuret reagent remains blue.
  • Positive Result (Presence of Peptide Bonds): If peptide bonds are present, the blue Biuret reagent turns violet or purple, indicating the presence of proteins or peptides.

    The intensity of the violet or purple color can be correlated with the concentration of proteins in the solution. The more peptide bonds present, the more intense the color change.
